Polyclonal immunoglobulin (Ig) G autoantibodies against insulin have been
identified in sera of healthy cats. We purified and fractionated insulin-binding IgGs from
cat sera by affinity chromatography and analyzed affinity of insulin-binding IgGs for
insulin and their epitopes. Following the passing of fraction A, which did not bind to
insulin, insulin-binding IgGs were eluted into two fractions, B and C, by affinity
chromatography using a column fixed with bovine insulin. Dissociation constant (KD) values
between insulin-binding IgGs and insulin, determined by surface plasmon resonance analysis
(Biacore™system), were 1.64e−4 M for fraction B (low affinity IgGs) and
2e−5 M for fraction C (high affinity IgGs). Epitope analysis was conducted
using 16 peptide fragments synthesized in concord with the amino acid sequence of feline
insulin by an enzyme-linked immunosorbent assay. Fractions B and C showed higher
absorbance (affinity) of the peptide fragment of 10 amino acid residues at the
carboxyl-terminal of the B chain (peptide No. 19), followed by peptide fragments of 6 to
15 amino acid residues of the B chain (peptide No. 8). Fraction C showed a higher
absorbance to 7 to 16 amino acid residues of the B chain (peptide No. 5) compared with the
absorbance of fraction B. Polyclonal insulin-binding IgGs may form a macromolecule complex
with insulin through the multiple affinity sites of IgG molecules. Feline insulin-binding

Two anti-inflammatory omega-3 polyunsaturated fatty acids (PUFAs) of stearidonic acid (SA) and eicosapentaenoic acid (EPA) and one pro-inflammatory omega-6 PUFA of arachidonic acid (AA) were isolated from the edible brown seaweed Undaria pinnatifida. SA was active against mouse ear inflammation induced by phorbol myristate acetate, with IC50 values of 160, 314, and 235 microg per ear for edema, erythema, and blood flow, respectively. EPA was also active against edema, erythema, and blood flow, with IC50 values of 230, 462, and 236 microg per ear, respectively. Although AA at low concentrations showed anti-inflammatory activities when measured 10 h later, AA doses of more than 243 microg per ear induced inflammatory symptoms 1 h later. Mature thalli generally had larger amounts of PUFAs than young thalli. The algal blade contained more omega-3 PUFAs than were found in other parts, while the holdfast contained extremely high amounts of AA. The objective of this study was to examine the health and meat production of cloned sows
and their progenies in order to demonstrate the application of somatic cell cloning to the
pig industry. This study compared the growth, reproductive performance, carcass
characteristics and meat quality of Landrace cloned sows, F1 progenies and F2 progenies.
We measured their body weight, growth rate and feed conversion and performed a
pathological analysis of their anatomy to detect abnormalities. Three of the five cloned
pigs were used for a growth test. Cloned pigs grew normally and had characteristics
similar to those of the control purebred Landrace pigs. Two cloned gilts were bred with a
Landrace boar and used for a progeny test. F1 progenies had characteristics similar to
those of the controls. Two of the F1 progeny gilts were bred with a Duroc or Large White
boar and used for the progeny test. F2 progenies grew normally. There were no biological
differences in growth, carcass characteristics and amino acid composition among cloned
sows, F1 progenies, F2 progenies and conventional pigs. The cloned sows and F1 progenies
showed normal reproductive performance. No specific abnormalities were observed by
pathological analysis, with the exception of periarteritis in the F1 progenies. All pigs
had a normal karyotype. These results demonstrate that cloned female pigs and their
progenies have similar growth, reproductive performance and carcass quality
characteristics and that somatic cell cloning could be a useful technique for conserving

ABSTRACT: Stocking of exogenous, hatchery-reared white-spotted charr Salvelinus leucomaenis has been conducted throughout much of their range in Honshu Island, Japan, to increase angling opportunities. Although the native charr populations are thought to have declined because of hybridization with introduced fish, their distribution and genetic status have been uncertain. Fine population structures of charr in the upper Tone River drainage were examined using mitochondrial DNA and microsatellite analyses so as to clarify the presence of native populations. One common mtDNA haplotype was detected in all populations in the Ohashi River and Watarase River, and four and one tributary populations were monomorphic for such haplotypes, respectively. However, several haplotypes, considered to have originated from stocked hatchery fish, were observed in the stocked and the remaining populations. Judging from the genetic integrity over a fine geographic scale, the former were considered as indicative of native populations and the latter as admixtures with hatchery fish. Comparisons of genetic diversity, deviations from the Hardy–Weinberg equilibrium, principal component analysis, and relatedness estimations based on microsatellite DNA can also provide evidence for distinguishing native populations from those influenced by hatchery fish. 相似文献

In the wheat (Triticum aestivum L.) cultivar ‘Zenkoujikomugi’, a single nucleotide polymorphism (SNP) in the promoter of MOTHER OF FT AND TFL1 on chromosome 3A (MFT-3A) causes an increase in the level of gene expression, resulting in strong grain dormancy. We used a DNA marker to detect the ‘Zenkoujikomugi’-type (Zen-type) SNP and examined the genotype of MFT-3A in Japanese wheat varieties, and we found that 169 of 324 varieties carry the Zen-type SNP. In Japanese commercial varieties, the frequency of the Zen-type SNP was remarkably high in the southern part of Japan, but low in the northern part. To examine the relationship between MFT-3A genotype and grain dormancy, we performed a germination assay in three wheat-growing seasons. On average, the varieties carrying the Zen-type SNP showed stronger grain dormancy than the varieties carrying the non-Zen-type SNP. Among commercial cultivars, ‘Iwainodaichi’ (Kyushu), ‘Junreikomugi’ (Kinki-Chugoku-Shikoku), ‘Kinuhime’ (Kanto-Tokai), ‘Nebarigoshi’ (Tohoku-Hokuriku), and ‘Kitamoe’ (Hokkaido) showed the strongest grain dormancy in each geographical group, and all these varieties, except for ‘Kitamoe’, were found to carry the Zen-type SNP.
